We are sequencing Treponema pallidum DNA isolated from subjects enrolled from geographically diverse global sites as part of an NIH/NIAID Cooperative Agreement to support syphilis vaccine development (U19AI144177) and a complementary Bill and Melinda Gates Foundation-supported syphilis genetic epidemiology project. Most samples were directly sequenced from genital ulcer chancre swabs or skin biopsies. Whole-genome sequencing was performed using Illumina sequencing after T. pallidum enrichment using Agilent RNA baits +/- pooled, parallel whole-genome amplification.
